About MOTS-c

MOTS-c, the mitochondrial open reading frame of the 12S rRNA type-c, is a 16-amino-acid peptide, Met-Arg-Trp-Gln-Glu-Met-Gly-Tyr-Ile-Phe-Tyr-Pro-Arg-Lys-Leu-Arg, encoded by a short open reading frame within the mitochondrial 12S ribosomal RNA gene. It was described in 2015 by Changhan Lee, Pinchas Cohen and colleagues at the University of Southern California.

Together with humanin, it is one of the best-studied mitochondrial-derived peptides: short peptides encoded in mitochondrial DNA that act as signals to the rest of the cell.

Research context

The original work reported that MOTS-c inhibits the folate cycle and de novo purine synthesis, leading to accumulation of the AMP mimic AICAR ribonucleotide and activation of AMPK. Later studies described its translocation to the nucleus under metabolic stress, where it influences gene expression.

It is used in research on skeletal-muscle glucose metabolism, AMPK signaling and physical capacity in aging mouse models.

How does MOTS-c relate to AICAR?

By inhibiting the folate cycle, MOTS-c causes the cell's own AICAR ribonucleotide (ZMP) to accumulate, which activates AMPK. The AICAR listed separately is the cell-permeable riboside that cells convert into the same ribonucleotide.

Why does the supplier sheet label these codes GHK-CU?
