About 5-Amino-1MQ
5-Amino-1MQ is 5-amino-1-methylquinolinium, a small quinolinium compound rather than a peptide. It inhibits nicotinamide N-methyltransferase (NNMT), a cytosolic enzyme that transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide, producing 1-methylnicotinamide (1-MNA).
It comes from a structure-based series of methylquinolinium NNMT inhibitors developed at the University of Texas Medical Branch and described in 2018 as membrane-permeable and relatively selective.
Research context
Because NNMT consumes nicotinamide and SAM, inhibiting it is a way to study links between the NAD+ salvage pathway, cellular methylation potential and adipocyte metabolism. In cultured adipocytes, published work reported lower 1-MNA, higher NAD+ and SAM, and reduced lipogenesis; high-fat-diet mouse studies examined adipose tissue mass and metabolic parameters.
