Tesamorelin keeps the complete 44-amino-acid sequence of human growth hormone-releasing hormone and adds a trans-3-hexenoyl group to the N-terminal tyrosine. This single modification protects it from DPP-4 cleavage, the main route by which native GHRH is inactivated.
It therefore sits between the short GHRH(1-29) analogues, sermorelin and CJC-1295, and the native hormone: full length like GHRH, but stabilized.
