About FOXO4-DRI
In senescent cells, the transcription factor FOXO4 binds p53 and holds it in the nucleus, which helps those cells avoid apoptosis. FOXO4-DRI is a peptide designed to compete with that interaction. It is built from D-amino acids in reversed sequence order, the D-retro-inverso strategy, which mimics the side-chain layout of the natural sequence while resisting proteases.
The peptide was described in a 2017 study by Baar and colleagues, which reported selective apoptosis of senescent cells in culture and effects in mouse models of accelerated and natural aging.
