About Semaglutide
Semaglutide is an analogue of human glucagon-like peptide-1, GLP-1(7-37). Two amino-acid substitutions, α-aminoisobutyric acid (Aib) at position 8 and arginine at position 34, protect it from cleavage by dipeptidyl peptidase-4 (DPP-4) and allow a C18 fatty diacid side chain to be attached through a linker at lysine 26.
The fatty diacid binds albumin reversibly, which is the main reason semaglutide lasts far longer than native GLP-1. It is a selective GLP-1 receptor agonist and is widely used as a reference compound in GLP-1 receptor pharmacology.
Research context
Semaglutide is used to characterize GLP-1 receptor activation, receptor internalization and downstream cAMP signaling, and in preclinical studies of glucose homeostasis, gastric emptying and central appetite circuits.
Because its structure and pharmacology are well documented, it is also the usual comparator for newer GLP-1, dual and triple agonists, and it is one of the two components of the CagriSema combination.
