Liraglutide is an analogue of human GLP-1(7-37) with a single substitution, arginine for lysine at position 34, and a C16 palmitic acid attached to lysine 26 through a glutamic acid spacer. It shares about 97% of its sequence with native GLP-1.
The fatty-acid chain promotes self-association and albumin binding, extending the half-life compared with native GLP-1. Liraglutide was one of the first acylated GLP-1 analogues and preceded semaglutide, which builds on the same acylation strategy.
